CaloPix Connect: An Open API to Integrate AI into the Digital Pathology Workflow

Artificial Intelligence is rapidly transforming pathology. Automated metastasis detection, biomarker quantification, scoring assistance, therapeutic prediction—the use cases are multiplying and paving the way for faster, more precise diagnoses.

Yet one major challenge remains: integrating these AI solutions seamlessly, securely, and sustainably into the daily operations of laboratories and pathology departments. To address these challenges, Tribun Health designed CaloPix Connect, a generic API built to easily connect third-party AI solutions to CaloPix®, our image management system where pathologists view digital slides and perform their diagnoses. CaloPix Connect acts as an interoperability layer, ensuring smooth, secure integration that meets the strict requirements of medical environments.

The AI pathology market is highly dynamic, with many vendors offering algorithms that may be complementary—or competing. In this context, laboratories need flexibility: the ability to freely select the tools that best fit their clinical practice, without being locked into a closed ecosystem.  

With CaloPix Connect, Tribun Health has chosen openness and interoperability:

AI accessible directly within CaloPix®, without disrupting daily practice 

Artificial Intelligence only creates real value when it integrates naturally into everyday practice. The goal of CaloPix Connect is simple: to allow medical teams to leverage the power of AI without having to change their existing workflows or daily routines.

In practice, thanks to CaloPix Connect, AI tools appear directly within the CaloPix® interface, as fully integrated components of the pathologist’s workstation. With a single click, an analysis can be launched directly from the digital slide: the image is automatically pseudonymized, securely transmitted to the AI provider, and then processed.

 

The progress of the analysis can be monitored in real time, and the main results (scores, classifications, counts, etc.) are displayed directly in the CaloPix® viewer. Depending on the solution, PDF reports or contextual links to the provider’s viewer allow users to explore the analysis in greater depth. 

The result: a seamless experience without multiplying tools, preserving pathologist’s focus, improving efficiency, and reinforcing the clinical value of AI in diagnosis.

 

Simplified, secure and scalable technical integration

Behind this apparent simplicity lies an architecture in which CaloPix Connect plays a central role. This API was designed to meet the technical, regulatory, and organizational requirements of healthcare institutions, while preserving the stability of CaloPix®.

Each AI provider connects to CaloPix Connect through a simple configuration file, detailing their algorithms, supported image formats, authentication methods, and results structure. CaloPix® remains the single point of entry for users, while CaloPix Connect orchestrates the technical exchanges in the background. It handles:

  • Unified authentication management
  • Systematic pseudonymization of data in compliance with GDPR
  • Secure transfer of images to cloud or on-premises infrastructures
  • Automated retrieval and display of results
  • Data synchronization when results are updated within the AI provider’s environment

This standardization drastically reduces integration time, maintenance costs, and operational complexity, while ensuring a high level of security, traceability, and compliance.

Qritive – biomarkers and oncology decision support
QAi Lymph Node Dx is designed to detect metastases in lymph nodes with high accuracy. By analyzing the entire digital slide, the AI highlights suspicious regions that may contain macro- or micrometastases. It acts as a safety net for pathologists, reducing the risk of missed diagnoses while improving both diagnostic sensitivity and workflow efficiency.

Aiforia – automated tissue analysis
Aiforia Prostate supports multiple key steps in prostate cancer diagnosis on biopsy slides. It automatically detects suspicious tumor regions, including small, hard-to-spot foci. The solution also assists with Gleason grading by analyzing tumor architecture and suggesting patterns, while providing automated tumor quantification. The result is improved diagnostic accuracy, greater consistency, and increased efficiency in routine practice.

DeepPath – automated metastasis detection
Lydia is an AI tool focused on detecting metastases in lymph nodes—an essential but time-consuming task. It identifies and highlights suspicious areas, including micrometastases and isolated tumor cells. Acting as a second reader, it helps pathologists focus on the most relevant regions, improving both detection sensitivity and overall efficiency.

Primaa – specialized solutions in breast and dermatopathology

  • Cleo Breast supports the interpretation of breast biopsies by detecting and classifying lesions, while highlighting relevant regions. This improves diagnostic consistency and confidence, while streamlining workflow in high-volume environments.
  • Cleo Skin analyzes the morphology of skin lesions on digital slides. Given the variability of dermatopathology cases, it helps distinguish benign from malignant conditions, including melanoma, by detecting suspicious patterns and assisting classification. This leads to improved reproducibility and efficiency in routine practice.

Owkin – theranostic AI
Owkin develops advanced AI solutions for diagnostic support and therapeutic prediction, enabling a more personalized approach to patient care.

  • MSIntuit is designed to optimize MSI testing in colorectal cancer by analyzing routine H&E slides. It predicts microsatellite stability directly from histology, helping reduce the need for systematic molecular testing. By prioritizing confirmatory analyses and avoiding unnecessary tests, it improves efficiency while supporting the identification of patients eligible for immunotherapy.

  • Ralps Risk BC focuses on early-stage breast cancer by assessing the risk of recurrence from standard histology slides. By extracting subtle morphological features beyond traditional clinical and molecular markers, it helps clinicians refine patient stratification and adapt treatment decisions, particularly regarding chemotherapy. This contributes to a more personalized and accessible care pathway.

  • TLS Detect enables the identification and quantification of tertiary lymphoid structures directly on digital pathology slides. As these immune structures are associated with response to immunotherapy but remain difficult to assess consistently, the solution provides a standardized and objective evaluation of the tumor immune environment. It supports more reliable patient stratification and opens new opportunities in immuno-oncology research.

Ibex – AI-powered cancer diagnostics
Ibex develops AI solutions designed to support pathologists in cancer diagnosis by improving accuracy, consistency, and efficiency across routine workflows.

  • Ibex Prostate is designed to assist in prostate cancer diagnosis on biopsy slides. It automatically identifies and highlights suspicious regions, including small and difficult-to-detect tumor foci. Acting as a second reader, it helps reduce oversight risk while improving diagnostic confidence and workflow efficiency.

  • Ibex Breast focuses on breast cancer by detecting and classifying malignant and pre-malignant lesions on histology slides. It highlights relevant regions and supports more consistent interpretation, helping pathologists manage high case volumes while improving diagnostic accuracy and reproducibility.

  • Ibex Gastric supports the detection of gastric cancer and pre-cancerous lesions. By analyzing entire digital slides, it identifies suspicious areas that may be challenging to detect, particularly in early-stage disease. This enhances sensitivity, supports early diagnosis, and contributes to more efficient routine practice.
